Research Abstract |
Rho-kinase, an effector molecule of Rho gene, may reduce cellular adhesive potential (indispensable for cancer invasion and matastasis) and stimulate cancer's migrating potentials. The functions and actionmechanisms of Rho-kinase have been attracting close attention. Wepreviously reported that Rho-kinase and myosin phosphatase are involved in the regulation of microtubules as well as actomyosin thread regulation. To date, hardly any studies have been published concerning metastasis of human oral carcinoma cells. In our previous studies, of oncogenes is in nude mice with oral carcinoma cells implanted on their backs, cancer cell matastasis to other organs (including lymph nodes) was not observed. To study metastasis of oral carcinoma, it is necessary to develop a model of metastasis allowing a comparison between cancer cell lines with metastasizing potentials and those having oncogenic potentials but no metastasizing potentials. A plasmid inducing high Rho-kinase expression was introduced to primary squamous cell carcinoma cell line NOS-1 of mandibular gingiva to examine the possibility of isolating tumor cell lines with high metastatic potentials by reinforcing malignancy level and migration potentials. We obtained stable transfectants permanently expressing active type Rho-kinase. When measured by the gold colloid method. the migration potentials of the clones of these transfectants was about twice as great as that of the control cells. This indicates enhanced migration potentials of the clones expressing active type Rho-kinase. In orthotopic implantation of these transfectants to the nude mouse tongue, tumor take and oncogenic potentials were noted. but no matastasis to lymph nodes or other organs was observed.
