Research Abstract

古細菌rRNA遺伝子には多数の可動性イントロンが散在し,その各々の内部ORFにはLAGLIDADG型ホーミングエンドヌクレアーゼ(HEase)がコードされている.HEaseはこれらのイントロンの部位特異的転移を司る鍵酵素であり,ゲノム塩基配列の中からイントロンの転移標的を検索・切断しホーミング(homing)過程を開始させる役割を担う.またLAGLIDADG型HEaseは14-40bpの長いDNA配列を認識するレアカッター酵素であることもこれまでの我々の研究から明らかになっている.本年度は,広義の「制限酵素」の範疇に属するLAGLIDADG型HEaseの構造と機能について解析した.
海洋性超好熱古細菌Aeropyrum pernixおよび陸上温泉から分離された超好熱古細菌Pyrobaculum oguniense, Thermoproteus spp.から19種の新規な認識配列をもつ酵素を同定し,I-ApeI,I-ApeII,I-Tsp061I等と名づけた.いずれも14〜22bpの長さの偽回文配列または非回文配列を認識するレアカッター酵素であり,それらの基質特異性を詳細に検討した.I-Tsp061Iの結晶については分解能2.0Åの精密立体構造を解明することに成功し,その原子座標をPDBに登録した(PDB ID : 1VAW).I-ApeII遺伝子にランダム変異を導入することにより,その基質特異性が野生型とは異なる人工レアカッター酵素の創製に成功した.立体構造のモデリングから,この変異体における基質特異性の変化は,標的DNAとの相互作用を司るDNA結合表面(βサドル構造)の柔軟性が増強されたことによるものと推測された.
