Research Abstract

本研究は、宇宙フライトラットの骨格筋をDNAマイクロアレイ法で解析し19倍に増加を認めたオステオアクチビン遺伝子について検討を行った。そこで、我々はオステオアクチビンの機能を解明するためにトランスジェニックマウスを作製した。このトランスジェニックマウスを筋肉や骨の萎縮をきたす坐骨神経切除に供すると、Wild-typeマウスに比べ萎縮の程度が低かった(約8%)。さらに、坐骨神経切除したトランスジェニックマウスの骨格筋においては萎縮率の軽減に伴い、MMP-3の発現が著明であった。オステオアクチビンのトランスジェニックマウスにおいて坐骨神経切除による筋萎縮が軽度であったのは、細胞外マトリックスの構築と修復に関係のあるMMP-3を介している可能性が考えられた。
また、無重力による骨萎縮に関する2回目の宇宙実験(STS-107)を本研究と並行して行う予定であったが、スペースシャトル「コロンビア号」の事故によりその実験は頓挫した。しかし、宇宙航空研究開発機構(JAXA)を通してSTS-58の骨のサンプルを今年の3月に入手できることになったので当初の予定通り骨サンプルを用いた実験を行っていく予定である。
