自律的な分子分離に向けた微小管運動の電気的制御による分子設計論の確立

Outline of Annual Research Achievements

平成28年度は,当初の目的通り,高精度な微小管分離システムを実証した.まず,これまで提案してきた微小管の特性制御技術を応用し,キネシン上を運動する微小管を複数方向に輸送する技術を確立した.微小管の重合条件の調整,および微小管へのDNAの修飾により,曲げ剛性と電気泳動移動度を改変した複数の微小管を作製した.それぞれの一様電界中における運動方向を定量的に評価した後,その結果を基に分離に最適なマイクロ流体デバイスを設計した.微細加工技術を用いて作製したマイクロ流体デバイス内で,特性の異なる2種類の微小管を運動させた結果,一様なアッセイ環境下においても微小管を約80%の精度で分離できることを初めて実証した.以上の成果は,キネシンと微小管の系を用いた高次機能を有する分子システムの構築につながるものであり,マイクロナノシステムに関する国際会議MicroTASにおいて口頭発表で採択された.さらに,CBMSによる旅費助成を受けるなど,世界的に認められつつある.
また,平成28年度においては,生物物理学的な方面にも研究を発展させ,微小管の曲げ剛性に関する新たな知見を得た.従来の曲げ剛性の測定値には2桁程度のばらつきがあるなど,曲げ剛性に関する統一された見解はなかった.画像解析における微小管の位置決定精度,および微小管の伸長速度を変えて曲げ剛性を測定し,これまで考慮されてこなかったこれらの条件が曲げ剛性を決定する重要な要因であることを明らかにした.さらに,曲げ剛性の正確な測定手法を確立することで,微小管の伸長速度がある閾値を超えると,曲げ剛性が急速に低下するという新たな知見を得た.以上の成果は,曲げ剛性の関わる細胞内輸送の制御機構の解明につながるものであり,生物物理学に関する国際会議Biophysical Societyに採択され,分野を跨いで注目されている.
