Outline of Annual Research Achievements

1 クワガタムシ科における微生物共生関係の解明
クワガタムシ科の共生酵母は，雌成虫の菌嚢を介して次世代に伝達され，幼虫が食べる木質バイオマスの消化に密接に関連していると考えられる．本研究では、日本産クワガタムシ科の約40種のうち一部の離島固有種を除いた計32種について，成虫の菌嚢または幼虫消化管に存在する共生酵母を解析・同定した．クワガタムシ科の共生酵母の大半はScheffersonyces属のキシロース醗酵性酵母であったが、特殊な生態を持ついくつかの希少種クワガタムシからは、これまでに知られていない全く新規の酵母群が発見された。
また，酵母以外の微生物との共生についても新規の知見が得られた。例えば、フィリピンに生息するクーランネブトクワガタの成虫腸管からはツリガネムシ類に近縁な繊毛虫が見出された。陸上生物に繊毛虫が共生あるいは寄生する現象は大変珍しく、この発見については当年度内に論文化をおこない、Zoological Science誌に受理された。
2 共生酵母の地理的分化と遺伝的多様性
クワガタムシ共生酵母の大半を占めるScheffersomyces属酵母について、詳細な系統解析を行った。これまで酵母の系統解析に汎用されてきた26SリボソームRNA遺伝子やITS領域はScheffersomyces属内の変異が少なく，共生酵母とホスト昆虫の間の種特異性や，酵母の地理的分化などを調べるためには，これらに替わる新たな分子マーカーを開発する必要があった．そこで，日本産と韓国産のルリクワガタ属の共生酵母を対象として，東京大学および韓国の研究者と共同で，核リボソーム遺伝子のIGS領域における分子マーカーを開発し，その有用性について検討した．また、開発した分子マーカーを用いて、東アジアのルリクワガタ属共生酵母の遺伝的分化について調べた。これらの内容については論文査読中である。
