Research Abstract

「疎水性」の分子起源を分子性流体用積分方程式論に基づいて解析した。メタンのような疎水性溶質の溶解度が70-80℃で最小値をとるという挙動は,適当にモデル化した単純溶媒でも再現できる。水の特性の多くは,水素結合そのものや回転運動などにあるのではなく,小さな分子サイズと強い引力相互作用にある。溶質近傍における水分子間水素結合の強化(構造化)は,極めて低温で大きな溶質の場合を除けば,それによるエントロピー損失がエンタルピー利得を大きく上回るために好ましくない。
蛋白質は,高圧下では,水のエントロピーを増加させるために,内部に水分子が1分子層程度侵入したようなスウェリング構造に変性する。蛋白質の水和エントロピーを
(1)蛋白質-水間の2体相関レベルの水和エントロピーのうち,排除容積EVに依存する成分(朝倉-大沢理論で計算した水和エントロピー),
(2)それのうち,EV以外に依存する成分,
(3)蛋白質-水-水間の3体相関レベルおよび多体相関レベルの水和エントロピー
に分解して解析した。さらに,(2)と(3)の各々を水分子の並進運動の制限からの寄与と回転運動の制限からの寄与に分解した。その結果,高圧下で変性した場合に水のエントロピーが増加する原因は,(3)の並進運動の制限からの寄与にあることが分かった。(1)と(2)においてはいずれも損をする。内部に浸透した水分子の並進運動と回転運動はより制限されるものの,蛋白質外部における水分子の混み合いが大きく緩和されるのである。
分子性流体用積分方程式論と形態熱力学を統合した独特の理論を用いて,蛋白質の水和の熱力学量を高精度でかつ高速で計算できる新しいアプローチを開発した。このアプローチに基づいて構築した自由エネルギー関数を用いて,合計15種類の蛋白質に対し,各々天然構造と600-700通りのデコイ(偽物)構造の中から天然構造を射当てることに成功した。
