Research Abstract |
Adiponectin, which is secreted specifically by adipose tissue, has been shown to have an anti-atherosclerotic effect and to improve insulin resistance. The aims of this study were to determine the correlations of plasma adiponectin concentration with insulin resistance, atherosclerosis and its risk factors and to determine the association between aging and adiponectin level from the aspect of influence of renal function and sex hormones in humans. The subjects were 383 male inhabitants in two rural communities in Hokkaido, Japan. A negative correlation was found between homeostasis model assessment (HOMA) as an index of insulin resistance and adiponectin concentration. hCRP and pulse wave velocity (PWV) were used as an index of atherosclerosis. Results of stepwise regression analysis for adiponectin showed that age, HOMA and serum triglyceride (TG) were independently correlated. with adiponectin. A significant negative correlation was found between adiponectin and hCRP in all subjects, and a significant negative correlation between adiponectin and PWV was also found. Serum adiponectin and blood urea nitrogen (BUN) levels were measured in 964 subjects aged 60ア 13 years. Testosterone and free testosterone levels were measured in 123 males, and estrone and estradiol levels were measured in 114 females. Adiponectin level increased linearly with aging in males, while it increased dramatically in females until their 50s. The patterns of changes in adiponectin were similar to those in BUN. In multiple regression analysis using adiponectin as a dependent variable, BUN was selected as a significant independent variable in subjects, while bioactive sex hormones were not selected as significant independent variables. A decrease in adiponectin clearance in the kidney may cause high levels of adiponectin in the elderly. Adiponectin level seems to be influenced more strongly by BUN than by sex hormones and to be increased by a decline in renal function with aging.
