Research Abstract |
Since fertile hybrids were obtained between indica and japonica rice, utilization of the enhanced heterosis has been attempted in the inter-subspecific hybrid But the genetic basis for the heterosis has not been adequately analyzed. To specifically compare the effect of heterozygotes and homozygotes at each quantitative trait locus (QTL) in indica-japonica hybrids, two backcrossed populations(BC_1F_1), indica/japonica//japonica and indica/japonica//indica, were produced and mapped with a set of simple sequence repeat (SSR) markers. Several QTLs for grain shape traits, seedling characteristics and leaf senescence were found in the BC_1F_1orBC_1F_2lines with a LOD score of 2.5 as threshold. The effects of genotypes on respective traits were analyzed at these QTLs to analyze genetic basis of the heterosis. QTLs were detected by comparing two marker genotypes (AA and Aa, or Aa and aa) or their progeny lines (BC_1F_2) in each of the backcrossed populations. The genotypes that were determined to be heterozygous in the BC_1F_1 lines might therefore have contained three genotypes in the BC_1F_2 lines with the predicted frequencies : (1/4AA+1/2Aa+1/4aa). No QTL was identified at which the heterozygotes (Aa) showed a higher value than the homozygotes (AA or aa) in the two-paired populations. This provides evidence that the two populations gave consistent phenotypic values in the measured traits following a linear order of genetic effects : AA>Aa>aa or AA<Aa<aa. These findings also suggest that no over-dominance (Aa>AA) was present. The examination of genotypic effect at each QTL in this study revealed that positive effects were contributed either by indica or japonica parents. In the F_1 genotype, the cumulative partial dominance effects of such heterozygotes are considered to give better performance than the parents.
