Research Abstract |
Staphylococcal Panton-Valentine leukocidin (PVL) is an important virulence factor, which causes leukocytolysis and tissue necrosis. PVL genes are highly associate with community-acquired methicillin-resistant S.aureus (CA-MRSA), which is appear to be worldwide emerging pathogen. We suggested that horizontal transmission of PVL genes have been occured by at least two types of PVL-carrying temperate bacteriophage among S.aureus. In this study, we determine the genome of several PVL-carrying phages and found the polymorphysm of PVL-carrying phage genomes. We also found the polymorphism of the orf containing attB core sequence for φPVL/φSLT. By using these results, we construct the PCR scan system for PVL-carrying phage typing which will become useful tool for the epidemiological study of PVL carrying S aureus strain including CA-MRSA. Coagulase typing of PVL-carrying strains was also performed and partial relationship between PVL-phage and coagulase types were observed. In addition, a novel structural protein ORF636 with 66 kDa of φSLT, which contains two 97-amino acid sequence repeats with the sequence GW in its C-terminal region, was identified as an essential structural protein for the adhesion of φSLT to S.aureus through staphylococcal lipoteichoic acid.
