Research Abstract |
Protein overexpression and gene amplification of the tyrosine kinase growth factor receptors such as ErbB2, epidermal growth factor receptor (EGFR) and Met by immunohistochemistry (IHC) and fluorescence in situ hybridization (FISH) respectively. The presence or absence of gene amplification of c-erbB-2 (chromosomal locus 17q11.2-q12), EGFR (7p12) and c-met (7q31) was determined using fluorescently labeled DNA probe sets purchased from Vysis, Inc. The results are as follows : Colorectal carcinomas Overexpression of ErbB-2 was found in 8 (3%) of 244 colorectal carcinomas and all of them had gene amplification. Overexpression of EGFR was observed in 19 (8%) of the 244 tumors, and gene amplification was found in 58% of them. Non-small cell lung cancers Overexpression of EGFR protein was found in 34% of 181 cases of non small cell lung cancers. Among these, EGFR amplification was demonstrated in 74%. sion. Bile duct carcinomas A total 221 biliary tract carcinomas consisting of 28 intrahepatic bile duct, 78 extrahepatic bile duct, 89 gallbladder and 26 ampulla of Vater were examined. Overexpression of ErbB-2 was found in 15.7%, 11.5%, and 5.1% of carcinomas of the gallbladder, ampulla of Vater, and extrahepatic bile duct, respectively, and 79% of them were associated with gene amplification. Overexpression of EGFR was found in 8.1% of tumors with no predominant location and also associated with gene amplification in high frequency (77%). Met overexpression, most frequent in intrahepatic bile duct carcinomas (21.4%), was not associated with gene amplification. Esophageal squamous cell carcinomas EGFR protein is overexpressed in 53 (50%) of the 106 esophageal squamous cell carcinomas examined EGFR gene amplification was observed in 15 of the 53 tumors, somewhat higher EGFR copy in 32 cases, and no increase in 6 cases.
