Research Abstract |
Aortic aneurysms are considered to be the remodeling of the aortic wall, involving fragmentation and decrease of elastic fibers in the tunica media. Matrix metalloproteinases (MMPs) have been implicated in collagen and elastin, which results in abdominal aortic aneurysm (AAA). Elastin is produced by smooth muscle cells (SMCs) and fibroblasts. Insulin-like growth factor-1 (IGF-1) is thought to regulate the expression of elastic gene. The aim of this study was to investigate elastic regeneration by IGF-1 gene transfection as a new strategy for aortic aneurysms. This gene therapy for aortic aneurysms may contribute to the development of new regenerative strategy for patients with AAA, especially for prevention of AAA rupture. Aortic SMC and rat aortic wall were transfected with IGF-1 as a preliminary study. After IGF-1 transfection elastic production will be assessed as a next step.
