Research Abstract

数ナノメートルから数十ナノメートルのナノ粒子表面に生体親和性の高いポリエチレングリコールをブラシ上に構築し、生体環境下で高い分散性と生体成分の非特異吸着抑制を達成させることに成功した。具体的には
i)CdS/ZnSe量子ドットの水溶化と分子認識の実現、さらには水中で安定に発光させることに成功した。また、蛍光エネルギー移動現象を利用し、新しいバイオセンシング技術を確立した。
ii)前年度調製したCHO-PEG/ポリカチオンとアニオン系リポソームのによって、分散安定化リポソームの調製に成功した。この複合体への薬物ローディングを行い、pHに応答して薬物リリースする新しいシステムの構築が確認された。
iii)フラーレン含有ナノ粒子のイメージング能の評価:前年度までに調製したガドリニウム内包フラーレンとCHO-PEG/ポリカチオンとの複合体の水の緩和時間に対する影響を検討し、イメージング剤としての有用性を示した。
iv)PEG/ポリカチオン・金コロイド複合体へのオリゴ核酸の担持:PEG/ポリカチオンを固定化した金コロイドの最表面のカチオンチャージを利用してオリゴ核酸を担持し、遺伝子キャリアへの展開を行ない、安定なDNAベクターの可能性を示した。
v)金コロイドに酵素を固定化し、PEGによる分散を試み、高い分散性でかつ耐熱性の高い酵素複合体の調製に成功した。
