Research Abstract

本研究目的は,従来のニトロイミダゾール型低酸素細胞放射線増感剤にp53を"一過的かつ可逆的"に阻害する機能を付加することにより低酸素腫瘍細胞の放射線感受性を高めつつ,腫瘍組織周辺に存在する正常細胞の細胞死(副作用)を低減するバイファンクショナル低酸素細胞放射線増感剤の開発である.本年度は,TX-2004,TX-2071及びTX-2072を用いて,野生型p53を組み込んだH1299/wtp53細胞と変異型p53を組み込んだH1299/mp53細胞に対する温熱死の防護効果を調べた.コントロール化合物としてpifithrin-αおよびpifithrin-βを用いた.その結果,H1299/wtp53細胞において,TX-2004はpifithrin-αおよびpifithrin-βと同等のp53依存的細胞死の防護が見られた.しかしながら,H1299/mp53細胞においても細胞死が見られたことから,p53特異的な作用機構であるとは決定できなかった.次に,更なる構造活性相関としてTX-2004のメチレンリンカーの長さを変化させたTX-2088(メチレン鎖=3)及びTX-2112(メチレン鎖=4)を合成し,同様にp53依存的細胞死の防護を調べた.その結果,H1299/wtp53細胞の温熱処理による細胞死に対して,TX-2088はTX-2004よりも高い防護効果を示した.よって,標的部位は不明であるが,イミノチアゾール骨格と2-ニトロイミダゾール骨格の間の距離にはある程度の自由度があり,メチレン鎖が3つのTX-2088の方がより空間的相互作用が有利に働くと考えられる.これらの結果は本研究にて初めて得られたものであり,本研究目的である「p53の阻害を介して正常細胞へのダメージを軽減する低酸素細胞放射線増感剤の開発」に対して極めて重要な知見であることが示唆された.
