Outline of Final Research Achievements |
I successfully established a technique recovering a pair of variable heavy and light antibody gene sequences from each B cell in human blood. This technique should be useful to identify autoantibody sequences against mutated cancer antigens. We showed that 2 days-polyclonal activation of B cell from human blood in cell culture followed by single cell sorting dramatically increased the rate of antibody gene recovery from a single cell in the next cDNA preparation and sequencing step. The high rate of recovery of antibody sequence makes this technique practical for obtaining antibody sequence from a limited number of B cell available in patients’ specimens and having their moderate-level of the specific binding affinity. This approach should be practical and applicable to obtain a small size of antibody library from a few patients’ specific B cells in human blood. Isolated antibodies by this method will be useful for identifying autoantigens including mutated cancer-related antigens.
