Outline of Annual Research Achievements

1. 両生類ツメガエル知覚神経系におけるガストリン放出ペプチド(GRP)系の組織化学解析
前年度行った遺伝子発現解析およびタンパク質発現解析の結果を受け、今年度は、中枢神経系におけるGRPの局在解析を行うため、成体ネッタイツメガエル(Silurana tropicalis)の脳および脊髄組織を用いて免疫組織化学解析を行った。その結果、脳－脊髄の広域にわたり、GRP免疫陽性を示す神経軸索が密に分布していた。さらに、延髄および脊髄において、GRP免疫陽性線維や細胞体が多数観察された。以上から、両生類ネッタイツメガエル知覚神経系においてもGRP系が発現していることが示唆された。
2. 硬骨魚類メダカ知覚神経系におけるGRP遺伝子、アミノ酸配列の同定
硬骨魚類メダカ(Oryzias latipes)のゲノム情報に基づいたバイオインフォマティックス解析によりGRP/GRP受容体のオ－ソログ遺伝子を単離した。得られたメダカGRP配列を既に報告されている他の脊椎動物におけるGRPホルモン前駆体の配列と多重配列アライメントを用いて解析した。その結果、今回同定したメダカのGRP配列もGRP受容体配列もこれまで報告された他の脊椎動物との高い相同性が示された。次に、GRPとGRP受容体の各臓器における発現分布をRT-PCRを用いて解析したところ、哺乳類と同様にメダカにおいてもGRPが中枢神経系と消化器系に発現し、GRP受容体は脳と脊髄で高発現することが明らかになった。また、ウエスタンブロット解析により、GRP受容体のタンパク質レベルでの発現を調べた結果、脳と脊髄において、メダカGRP受容体の予測される分子量に特異的なシグナルが確認された。以上から、メダカ脳・脊髄においてもGRP/GRP受容体ともに遺伝子およびタンパク質レベルで発現することを明らかにした。
