Outline of Final Research Achievements

Paracetamol (PARA) is an antipyretic and analgesic medicine widely used in many countries. However, the precise molecular targets of PARA and the mechanisms of antipyretic and analgesic effects by PARA have not been fully clarified yet.
The PARA-compound and Hela cell lysate were bound and reacted. A band of the proteins bound to PARA-immobilized beads was GSK-3β , which is involved in body temperature homeostasis. The protein was stained with the antibody of GSK-3β by Western Blotting. Luminescent signals, which is correlated with the amount of GSK-3β activity, significantly decreased with increasing dose of PARA. These results indicate that PARA binds GSK-3β and inhibits its activity at clinically relevant concentrations.

Academic Significance and Societal Importance of the Research Achievements

アセトアミノフェンの作用機序を解明するためにアセトアミノフェンが結合する標的タンパクを同定した結果,体温の恒常性に関与するGSK‐3βが同定された.アセトアミノフェンが有する解熱効果の一部はGSK-3βの阻害作用によるものである可能性が示唆された.本研究結果からGSK-3βを阻害することで新たな解熱薬が開発される可能性があるといえる
