細胞内侵入性細菌に対する細胞内認識システムとオートファジー誘導の解析

Research Abstract

咽頭炎、扁桃炎などの原因菌であるA群レンサ球菌の宿主細胞内侵入経路および、細胞内侵入後のA群レンサ球菌の細胞内動態につき検討を加えた。その結果、細胞の自己成分分解・再利用にのみ関与していると考えられていたオートファジーが、細胞内侵入細菌の駆逐に重要な役割を果たしていることが明らかとなった。A群レンサ球菌はエンドサイトーシス経路を通って細胞内に侵入する。侵入後、この細菌は内毒素SLOを使ってエンドサイトーシスによる分解を免れ細胞質に逃れる。細胞質に逃れたA群レンサ球菌はオートファジーにより捕捉され分解される。オートファジーミュータント細胞では、A群レンサ球菌が増殖し、再び細胞外に脱出することも確認された。これまでエンドサイトーシス経路が細胞内に侵入する菌を分解する唯一のメカニズムだと考えられていたが、本研究により細胞は細胞内に侵入した病原微生物をオートファジーによっても分解・除去できることが証明された。多様な細胞が殺菌に働く免疫システムである自然免疫の新しいメカニズムの発見として、今後種々の感染症の予防・治療法の開発に資するものと考えられる。
