Research Abstract |
We have previously reported the successful creation of tissue-engineered vascular structure and the 47 clinical implantation of these autologous tissue in the pulmonary circulation. Recently, several institutes started gene therapy and cell therapy in the clinical setting for the treatment of ischemic myocardial failure. In this study, we tried to establish the method to differentiate BMCs to myocardial phenotype in vitro, then implant to in vivo model. To obtain the beating cells in vitro from the GATA-4 transfected stormal cells in vitro, two differenct protocol was designed. In protocol A, the effects of liquid factors from neonatoal myocytes was examined in a co-culture dish. In protocol B, cell-to-cell interaction was examined using mixed culture system. The cell-sheet system will be applied to create adult cardiac cell sheet. Using a rat model, Dr Nadal-Ginard protocol (Mt Sainai, NY, USA) was applied and cultured stem cell will be grown in culture. After obtaining the sufficient number of cardiac stem cells, they will be implanted in a ischemic heart model of syngenic rat. All experiments are still underway.
