Research Abstract |
Staphylococcus hyicus is a causative agent of exudative epidermitis in pigs and its one of the products, such as S.hyicus exfoliative toxin (SHET) causes typical clinical signs of this disease. SHET possess the serine protease motif in its molecule, but not possess caseinolytic protease activity. Therefore target substance of SHET seems to be a specific protein. SHETA and SHETB bound to a 40 kDa-protein (P40) extracted from the skin of 1-day-old chicken. SHETA and SHETB bound to not only P40s from susceptible animals (1-day-old chicken and piglet) but also to P40 from non-susceptible animal (1-day-old mouse). Antisera to chicken P40 reacted with P40s from chicken, pig and mouse. However, SHETA and SHETB cleaved to chicken P40 and pig P40, but not cleaved to mouse P40. Amino terminal sequence of highly purified chicken P40 was almost same as amino acid sequences of extracellular domain of bovine, human, murine and canine desmoglein 1 (DSG1) and desmoglein 3 (DSG3). Then anti-human DSG1 serum and anti-human DSG3 serum were reacted with chicken P40, Pig P40 and mouse P40. Chicken P40, pig P40 and mouse P40 were reacted with anti-DSG1 serum, but not reacted with anti-DSG3 serum. After treatment of SHETA and SHETB, chicken P40 and pig P40 were not reacted with anti-DSG1 serum, while mouse P40 was still reacted with anti-DSG1 serum. From these findings, it is sugested that DSG1 is a target protein of SHET and that DSG1 from susceptible animal is cleaved by SHET, while DSG1 from non-susceptible animals is not cleaved.
