Research Abstract |
In this canine erythrocytes study, the effect of -196℃ or two step method(-196℃→80℃) cryopreservation was examined. Erythrocytes were mixed with cryoprotective solution containing hydroxyethyl starch (HES) or glycerol (GL). This mixture was filled into plastic bag made of vinyl chloride and frozen to -196℃ in liquid nitrogen. It was transferred to -80℃ deepfreezer and was preserved for 3 month. After thawing, erythrocytes were not washed, and autotransfused. 1)The canine erythrocytes frozen HES and GL at -196℃ could be autotransfused without side effects. The HES was considered to have a practical value rather than the GL. 2)HES was used a cryoprotective solution, the possibility of the plentiful cryopreservation of canine erythrocytes at two step method (-196℃→-80℃) was found out. 3)Canine erythrocytes were cryopreserved at -80℃ with HES for three months or more was found out. In recovery rate, osmotic fragility test and scanning electron microscopy, significant difference was not observed in each period. Therefore, -80℃ cryopreservation can preserve canine erythrocytes longer than liquefied preservation. 4)The canine erythrocytes frozen HES at two step method could be autotransfused without side effects. This two step method that could be autotransfused, and was considered to have a practical value in veterinary medicine.
