Research Abstract

RNA interference (RNAi) mediated by small interfering RNAs (siRNA) is a potent technology allowing the silencing of mammalian genes with great specificity. Technology that allows the downregulation of expression of specific genes expressed in the skin would have numerous application ranging from improving our understanding of basic biology of the skin to providing therapy for psoriasis vulgaris.
Angiogenesis is a prominent feature of a number of inflammatory human disease, including rheumatoid arthritis, psoriasis. Upreguration of vascular endothelial growth factor (VEGF) and placental growth factor (PIGF), a member of VEGF family, have been found in several conditions associated with pathological angiogenesis; however, its distinct role in the control of angiogenesis has remained unclear. To investigate the expression of VEGF and PIGF, the lesional skins of psoriasis patients were investigated with ELISA assay. We found strong upreguration of VEGF and PIGF protein levels in the lesional skin of psoriasis vulgaris patients, and designed siRNAs for VEGF or PIGF. Moreover, we test new vehicles for their accurate delivery.
