Research Abstract |
1.We have examined the antibodies to phosphatidylserine (PS), cardiolipin (CL), phosphatidylethanolamine (PE), and annexin A5 in patients with recurrent pregnancy loss, recurrent IVF-ET failure, preeclampsia and having IUGR fetus. From our research, we focused on IgG-isotype and purified IgG types of each antibody. 2.Follicular fluid in patients with recurrent IVF-ET failure had IgG types of anti-PS, CL, and PE antibodies, and we found the relationship between poor fertilization rate and IgG antibodies. 3.Annexin A5 was purified from the placenta and recombinant annexin A5 was also made. 4.We used several liposomes (PS/PC, CL/PC, PE/PC, PI/PC, SM/PC, PC only), platelets, trophoblasts as sources of phospholipids, and tested the relationship among annexin A5, anti-annexin A5 antibodies, phospholipids, antiphospholipid antibodies with ELISA、 Biacore、 Immunoblot, flowcytometry, and immunohistochemistry, which revealed annexin A5 can bind all of phospholipids but PC only. 5.We examined the effect of purified antibodies to the murine oocytes, sperms and embryos by micro- and macro-scopically. 6.Anti-laminin antibodies were related to pregnancy loss. 7.Peripheral NK cell activity was elevated in patients with infertility. 8.Laser scattered aggregometry system revealed that those antibodies made platelets aggregate. 9.All of above findings suggests that infertility and pregnancy loss are similar.
