Research Abstract |
Head investigator and investigators have provided the results from this grant projects. (1)One hundred and eighty one gingival crevicular fluids samples were obtained from 52 periodontitis patients and interleukin-1 (IL-1)β and soluble IL-1 receptor type 2 (sIL-1Rt2) were measured by using ELISA assay kits. The concentration of IL-1β and sIL-1t2 were positive correlated with the periodontal disease severity. On the other hand, GCF IL-1b and sIL-1t2 concentrations from chronic periodontitis (CP) patients and aggressive periodontitis (AgP) patients were also examined. IL-1β was no different between CP (82.2±12.3ng/ml)and AgP(68.3±9.4ng/ml), however, AgP sIL-1Rt2(3.0±0.6ng/ml) was significantly decreased compared with CP sIL-1Rt2(1.1±0.3ng/ml). Therefore, these results suggest that AgP disease character of rapid periodontal tissue breakdown may relate to the decreased sIL-1Rt2 production. rtment of Periodontology in Aichi-gakuin University from 2002 to 2004. Polymorphisms were identified by direct sequence method. After the single nucleotide polymorphisms (SNPs) identification, allelo- and haplotype analysis were performed. A 14.5-kb genomic region containing all exons and relevant exon-intron boundaries of IL1R1 and IL1R2 were sequenced in 24 AgPs and 24 controls. Within genomic regions containing all exons and relevant exon-intron boundaries in IL1R1 and IL1R2,38 SNPs were identified in 48 subjects. Five were located within coding elements, 2 in the 5' flanking region,22 within introns, and 9 in the 3' untranslated region. Frequency of 3 SNPs and 3 haplotypes in IL1R2 showed significant differences between patients with AgP and controls (p<0.05). (3)Gingival fibroblasts were isolated from chronic periodontitis patients. And cells were stimulated with Actinobacillus actinomycetemcomitans lipopolysaccharide, which is strongly associated with AgP. IL-1β and IL-1Rt2 mRNA gene expression were measured. However, protein production was only measured for IL-1β and not for sIL-1Rt2 in culture supernatant.
