Research Abstract |
本研究では、自然免疫系のアダプター分子MyD88の細胞機能の解明を目指し、MyD88に含まれるTIRドメインの立体構造及び機能の解析を行なった。 1)立体構造解析 溶液NMRで立体構造決定を行なうために、^<13>C,^<15>Nで標識したMyD88 TIRドメインを調製し、各種多核多次元核磁気共鳴(NMR)の測定を行ない、これらの測定データを基に^1H,^<13>C,^<15>N核の化学シフトの帰属作業、各種三次元NOESYの解析を行ない、二次構造要素の同定と^1H間の距離情報の取得を行なった。次に、これらのデータから、立体構造計算を行なった。得られたMyD88 TIRの立体構造は、既に構造が解かれているTLR1やTLR2のTIRドメインと類似しているが、ループ部分の長さや、一部のα-ヘリックスの配向などに差異があることがわかった。 2)機能解析 MyD88 TIRドメインの自然免疫シグナル伝達系における機能解析を行なうべく、HEK293培養細胞系でルシフェラーゼを使ったレポーターアッセイの系を立ち上げた。 コントロールのHEK293細胞では、IL-18で刺激することにより自然免疫系のシグナル伝達経路を通してNF-κBが活性化されることを確認した。その一方で、野生型のMyD88 TIRドメインを過剰発現させた場合は、そのDominant Negative効果により、IL-18刺激後のNF-κBの活性が有意に下がることも確認した。現在、立体構造情報に基づいて、種々のアミノ酸置換を導入したMyD88 TIRドメイン変異体をこの細胞に発現させて、IL-18刺激後にNF-κBの活性を測定し、どの残基がシグナル伝達に重要なのかを調べている。この他にRAW264.7細胞を使ってリポ多糖による刺激を行なうアッセイ系も確立した。
