Research Abstract

コオロギの初期胚発生メカニズムを,遺伝子機能解析により解明することを目的として研究を行った.昨年度確立した親導入RNAi法による遺伝子機能の系を用い,コオロギ初期胚発生に関与する遺伝子の機能解析を行った.その結果ショウジョウバエのギャップ遺伝子Kruppelのホモログがコオロギでもギャップ遺伝子として働くこと,ペア・ルール遺伝子even-skippedホモログがペア・ルール様の機能に加えてギャップ遺伝子の発現調節に関与することを見出した、また,コオロギのextradenticle遺伝子が,ショウジョウバエとは異なり顎領域の形成に必要であることを発見した.さらに,orthodenticle遺伝子がコオロギ胚の前方(頭部から胸部にかけて)の領域の形成に必要であることが明らかになった.加えて、これらの遺伝子の発現パターンについてもin situ hybridizationにより明らかにした.これらの成果と昨年度までの成果から,コオロギ前後軸パターン形成を司る遺伝子ネットワークのモデルを提唱するに至った.コオロギでは,ショウジョウバエのピコイドの代わりにcaudalとorthodenticleが前後軸パターン形成の初期に主要な役割を担っており,extradenticle, hunchback, Kruppelの順にギャップ遺伝子が活性化され,even-skippedもこの過程に関与していると考えられる.
