Outline of Annual Research Achievements

本研究「脳室帯のシステム科学的研究」は,脳細胞産生の場である「脳室帯」が異なる細胞群によって一種の「生態系」あるいは「社会」として構成されていることに注目し,大脳皮質細胞の「生産ライン」群がどのような近隣関係性をもって三次元的にパックされ,効率的細胞供給が果たされているか,マウスおよび他哺乳類を対象に明らかにすることで,大脳形成を支える原理,組織形成上の脆弱点,ヒトの先天性疾患の病因などを理解できるようになるとの期待のもと実施されている. 平成30年度は,これまでの成果を複数の論文として発表した.まず,新しく誕生した細胞が脳室面から速やかに立ち去り,次の細胞産生すなわち前駆細胞による分裂の邪魔にならないようにするために,弾性が利用されていることを見いだした.この弾性は,細胞の密集性により成立している.親細胞が分裂のため膨らむ際に周囲を押し,そこにいったん蓄えられた弾性エネルギーが,親細胞の分裂の直後に子細胞に戻ってきてその初動が助けられるという受動的な機構である(PLOS Biologyに報告).次に,そのような細胞密集による組織の弾性・圧縮性が一定の範囲内に限定されることに重要な意味がある(さもないと細胞づくりがうまく進まない)と見いだした(Developmentに報告).「限定」は,分化途上の細胞がお互いに絡み合ってバリア構造を作ることで果たされると分かった(煮物料理における落としぶたのような存在). さらに,脳原基での役割がまだよく分かっていないミクログリアに関して,脳室帯付近における振る舞いとそのしくみの一端を明らかにした(Genes CellsおよびeNeuroに報告).これらにより,脳室帯についての理解が大いに増しつつある.

Reason

地道に行なって来たイメージング,定量解析,力学的な試験などが集約されて大きな突破をいくつかの異なる切り口・対象において果たすことができた.こうした成果によって,新しい立ち位置・視点で残された課題に挑むことができる態勢が整いつつあると考えられ,最終年度の研究の進捗が大いに期待できる.

Strategy for Future Research Activity

脳室帯から発される放射状ファイバーは脳構造の支柱であるとともに,ニューロン移動のガイドに貢献することが知られる.このように「ニューロンを助ける」と知られてきた脳室帯由来ファイバーに対して胎生早期に誕生するニューロンが接線方向移動をすることで物理的な影響を及ぼし,放射状ファイバーの走行,ひいては大脳皮質形成,生後の領野マップ形成を下支えする可能性が生じてきた(昨年度,国際学会で部分的に発表)ので,イメージング,力学的シミュレーション,流れを止める・弱くする実験など,詰めの解析を行い,論文発表を目指す.
