Outline of Annual Research Achievements

内因性免疫とは、宿主へのウイルス感染に対して細胞自身が有する原始的免疫防御機構である。近年、「内因性免疫」はウイルス遺伝子のみならず宿主遺伝子にも変異を導入し発癌を促進する事が判明した。本研究では、このユニークな性質を持つ「内因性免疫」に着目し、1)ヒト乳頭腫ウイルス(HPV)による中咽頭癌発癌とEpstein-Barrウイルス(EBV)による上咽頭癌発癌を対比し解析する事で「内因性免疫」による頭頸部ウイルス発癌機構を解明すること、2)「内因性免疫」のregulatorであるエストロゲンとエストロゲン類似物質である内分泌撹乱物質による「内因性免疫」発現調節機構について解析し、ウイルス発癌に特異的な治療法
を開発することを目的とする。
本年度までに、中咽頭癌・上咽頭癌組織における性ホルモン受容体と内因性免疫の免疫組織学的解析およびmRNA発現解 析を行った。パイロットスタディの通り、HPV陽性中咽頭癌ではエストロゲン受容体の発現が強い傾向があった。またEBV陰性の上咽頭癌ではエストロゲン受容体が強く発現する傾向があった。又、内因性免疫因子の発現を免疫組織化学的に解析を行った。上咽頭癌においても中咽頭癌においてAPOBEC3A. APOBEC3G, AIDの発現がウィルス陽性であると強い傾向があることがわかった。また、宿主遺伝子、ウィルス遺伝子中の遺伝子変異量を3D-PCRおよびシークエンスにて解析を行ったが、いくつかの宿主遺伝子およびHPV遺伝子のE2領域、EBV遺伝子のLMP1領域で内因性免疫に特異的な遺伝子変異が検出された。そして、これらの事象を細胞株レベルでも、臨床検体から得られた知見と同様の結果が得られることが判明した。
これらの結果をもとに免疫不全マウスモデルを作成し、エストロゲンの効果によってこれらの癌細胞に効果があるのかを検討中である。
