Outline of Final Research Achievements

Vascular networks play pivotal roles in a variety of biological functions in the body. Blood vessels also contribute to cancer metastasis. This study aimed at understanding the mechanisms by which the vascular networks are patterned during early development using chicken embryos. Exploiting a flat structure of early embryo in which vascular morphogenesis proceeds, we have developed a method enabling visualization of individual endothelial cells during vascular remodeling. We have found that a subtle difference in blood flow results in a differentiation of disappearing vessels and thickening ones, leading to an overt patterning of vascular structures.

Academic Significance and Societal Importance of the Research Achievements

血管ネットワークは体の生命活動を支えるものであると同時に、ガン転移にも関与することから、血管ネットワーク、とくにその成立過程の理解は重要な課題と位置づけられる。血管形成過程では、①無秩序な構造をもつ原始血管網が作られ、②「血管リモデリング」によってダイナミックな再編成が起こり、③最終的に組織だった血管パターンが作られる。これまで②についてはよい解析系が無かったことからその理解が遅れていた。本研究ではこれらの課題を克服して血管リモデリングのしくみについて新規知見をえたことの意義は大きい。
