Research Abstract

The objective of this research is confirmations that quadrature fringes full field optical coherence tomography (QF IF OCT) proposed by us is valuable to study on neuro-activities in cortexes. It was impossible to obtain the en-face OCT images to show the neuro-activity in rat cortex. However we could notice the difference in depth intensity profiles through the comparisons between in vivo and in situ.
The measured three-dimensional (3D) area was 4 x 4 x 2.9 mm^3 (160x160x400pixels, XxYxdepth). In signal variations of OCT image on time, the mean of signal intensity in situ was 1.4 times larger than that of in vivo. The ratio of standard deviation to mean of signal intensity in vivo was 2 times larger than that of in situ. In vivo 3D imaging, the cross-section of thinned skull with thickness of 170 μm and the vein with of 150 μm were recognized without averaging. The vein with diameter of 70 μm was also recognized by averaging 300 en face images. In situ 3D imaging, cross-sections of the thinned skull and the vein with a diameter of 200 μm were measured. Compared the depth intensity profile in situ with in vivo, in case of in situ the maximum was observed below the surface at the depth of 1500 μm. This difference in intensity profiles between in vivo and in situ might be useful to measure and monitor the viability of cortexes non-invasively with time and spatial resolution.
