Research Abstract |
We investigated a transcriptome profile of mouse brain after hanging by serial analysis of gene expression (SAGE) method. RNA was extracted from the forebrain, hippocampus and cerebellum after hanging previously reported. Control groups were killed by decapitation without hanging. First, we investigated the level of immediate early genes (IEGs) mRNA such as c-fos, fos B and c-jun that induced another genes. As the results, the all IEGs drastically increased with hanging on all regions. These results indicated that transcriptome profiles might change with hanging. We have been able to obtain the transcriptome profiles after analyzing 12866 tags in hanging and 13334 tags in control. 7311 different genes in hanging and 7612 in control were observed. Compared with the two groups, 64 genes, including Ras homolog gene family member B and leucine-rich repeat neuronal 6A, were increased in hanging. On the other hand, 49 genes such as Kinesin family member C, and Phoshoprotein enriched in astrocytes 15 were decreased. These results contribute to clarify the pathophysiology of asphyxia in the lung and aid in the diagnosis of suffocation.
