Research Abstract |
Miniature swine are considered the most likely animals for xenoplantaion to human. However, their genome contains porcine endogenous retrovirus (PERV) capable of infecting human cells in vitro. In order to expole a potential use of miniature swine foe xenotransplantation, a new strain of Clawn miniature swine was developed in Kagoshima, Japan and investgatied for the presence of PERV genome. Peripheral blood mononuclear cells (PBMC) DNA of seven individual Clawn miniature swine was analyzed by the specific polymerase chain reaction (PCR) method, using the known primer for PERV gag, PERV pol, PERV-A env, PERV-B env, and PERV-C env. The PBMC DNA of three Large Yokshire pigs and three Landrace pigs as well as eight Kurobuta Berksire pigs, were examined for comparison. All of the tested samples were PERV-A, B, and C positive.
