Research Abstract

1. Aconitum Alkaloid
A new simultaneous determination method of aconitines and their metabolites from human body fluids such as blood or urine in case of aconite poisoning had been investigated by using solid phase extraction (SPE) and liquid chromatography - tandem mass spectrometry (LC-MS/MS). Six aconitines, well-known strongest neurotoxins, were isolated from domestic aconite roots in Japan. From these aconitines, six benzoylaconines, 8-hydroxy metabolites of aconitines, and four aconines, final 8,14-hydroxy metabolites of aconitines, were also synthesized. These compounds were used as standards. Various deuterated aconitines, benzoylaconines and aconines, and 8-alkoxyaconitines were also synthesized and examines as internal standards. By using our new method, all the alkaloids were rapidly determined at the levels of sub-ng/mL order, and this method seemed to be useful for forensic analysis or clinical analysis.
2. Veratrum Alkaloid
Our new SPE-LOMS/MS method was also applied for sim … More ultaneous determination of toxic and teratogenic Veratrum alkaloids from human body fluids such as blood or urine in case of Veratrum poisoning. Two juvenile alkaloids, four toxic and teratogenic alkaloids, and five strongly toxic ester-type alkaloids were isolated from Veratrum plants and used as standards. Methyllycaconitine was found to be the most suitable internal standard. By our new method, all the Veratrum alkaloids in blood or urine were also rapidly determined at the levels of ng/mL order, and this method was also thought to be useful for forensic analysis or clinical analysis.
3. Grayanotoxins
Another toxic plant Pieris japonica is also known as toxic from ancient time. We had experienced Pieris japonica poisoning case, and synthesized some toxic neurotoxin terpenoid grayanotoxins for standards and analyzed the raw plant, decoction residue and the vomit of the victim. From all the samples toxic grayanotoxins were determined, and we had found the enhancement mechanism of the toxic terpenoids in the decoction process or in the human gastric juices for the first time.
