Research Abstract |
(Introduction) Liver metastasis is a important predictive factor in colon cancer patients. When the mechanism is made clear, it will be possible for us to predict the high risk group of liver metastasis, and to develop immunotherapy. We investigated the role of CD4+CD25+Foxp3+ Tcell (regulatory T cell, reg T) in the formation of liver metastasis, using mice liver metastasis model. (Methods) 1, We made mice liver metastasis model, and tried to distinguish the highly metastatic cell from lower one. 2, Next, we analyzed the ratio of reg T and NK cells in peripheral lymphnodes and liver perfusate using above mentioned model. (Results) 1, Colon26, a mice colon cancer cell line, was injected into the spleen of the Balb/c mice. It was difficult to distinguish the highly metastatic cell from lower one. ells were injected into the spleen of Balb/c mice, and 4 days after injection, splenectomy was done, and 7and 10days after injection, the mice were sacrificed. In spleen preserved group, the mice were sacrificed 7and 10days after the injection of colon 26, without splenectomy. The ratio of NK cells in peripheral lymph nodes were significantly higher in splenectomy group both in 7 and 10 days after the injection(p<0.01). There were no significant difference in the ratio of regT. 3, We evaluated the changes of reg T and NK cells in mice after splenectomy, without the injection of colon26. Splenectomy decreased the ratio of both cells significantly (p<0.05). 4, It was difficult to evaluate the ratio of reg T and NK cells in liver lymphocyte, because we can't sort clearly the lymphocyte. 5, We made mice rectal cancer model and now are investigating the metastatic factor, for example MMP, and so on. (Conclusion) Splenectomy enhanced the liver metastasis in mice. The decrease ratio of NK cells in peripheral lymph nodes could enhance the liver metastasis, and the ratio of reg T did not affect the metastasis. Less
