Research Abstract

1.小口径血管再生のための新しい担体の開発:
Electrospinning法で作製したナノファイバーを用い、層状構造を呈した小口径血管再生用の担体を開発した。ラット頸動脈置換モデルを開発し、in vivo評価系を効率化した。PLA単独の担体は数日から1週間前後で破綻した。PCLの担体は生存するものもいた。
2.体制幹細胞から血管構成細胞への分化誘導法の開発(特に骨髄由来間葉系幹細胞の平滑筋細胞への分化誘導法および脱分化制御法)
A.サイトカイン・成長因子を用いた誘導法
骨髄間葉系幹細胞から平滑筋細胞への分化に、TGF-b1、アスコルビン酸の両者を添加することによって相加相乗的に平滑筋特異的蛋白や遺伝子の発現が亢進した。そのメカニズムにつき、細胞の分化・脱分化のキーレギュレーターであるシグナル伝達系の、AktやERKなどのプロテインキナーゼのリン酸化を検討したところ、ERKがアスコルビン酸単独投与よりも投与後30分の時点で有意に低くなった(脱分化の抑制が示唆された)。
B.メカニカルストレスの影響(バイオリアクターによる生理的ストレスと脱細胞化マトリクス用いた研究)
脱細胞化マトリクス(ブタ頸動脈)上に、磁性ナノ粒子を用いた層状細胞立体構築法で、骨髄間葉系幹細胞を層状に形成し、生理的ストレス環境下で培養したところ、平滑筋細胞特異的蛋白や遺伝子の発現が亢進した。同時に骨分化についても調べてみたところ、骨分化特異的遺伝子発現の上昇が認められた。このことは、すなわち、体制幹細胞を用いた血管再生において、細胞を血管構成細胞分化誘導させるとともに、細胞を目的外細胞への分化を抑制するような制御機構の開発も重要であることが示唆された。
