Research Abstract

申請者はこれまでに、Flt3-ITDが、下流のMEK/ERKなどのシグナル伝達系を活性化することにより、転写のco-repressorであるSMRT(silencing mediator of retinoic and thyroid hormone receptors)の核外排出をひきおこし、異常遺伝子発現制御、さらにはSMRTと相互作用する、AML細胞増殖抑制因子であるPLZF(promyelocytic leukemia zinc finger)の機能阻害によるAML細胞異常増殖に結びついていることを明らかにしている(Takahashi et al., 2004, Blood, 4650-4658)。このことは、Flt3変異型AML細胞において、SMRTは機能抑制状況下にあり、SMRT阻害剤である亜ヒ酸がFlt3変異型白血病に特異的な効果がある可能性を示している。
そこでFlt3阻害剤と亜ヒ酸の併用効果について、Flt3-ITD細胞を用いて検討を行ったところ、この2剤の併用がアポトーシスを強く引き起こすことが証明された。また、これら2剤投与における細胞増殖抑制効果の検討を行ったところ、Flt3-ITDをもつ細胞株(BaF3-Flt3-ITD,MV4-11,PL-21)ではChou talalay methodにおける細胞増殖阻害のcombination index(CI)が、2つの薬剤の全ての有効濃度で1.0以下(相加〜相乗作用)であったのに対し、Flt3野生型の細胞株(RS4-11,NB4)ではCIが1より大であった(拮抗作用)。以上よりFlt3変異をもつ白血病細胞に対して、Flt3阻害剤と亜ヒ酸の併用療法は極めて効果的であることが示された。
