Outline of Final Research Achievements

Eukaryotic translation initiation factor (eIF) 4A, a DEAD-box RNA-binding protein, plays an essential role in translation initiation. Two mammalian eIF4A paralogs, eIF4A1 and eIF4A2, have been assumed to be redundant because of their high homology, and the difference in their functions has been poorly understood. Here, combining multiple genome-wide methods, we found that eIF4A paralogs have preferential mRNA binders and that the bias originates from RNA binding protein LARP1, which binds to eIF4A1 selectively. Moreover, we revealed that eIF4A1 enhances the affinity between TOP mRNAs and LARP1 and thus ensures stronger translation repression of TOP mRNAs upon mTORC1 inhibition.

Academic Significance and Societal Importance of the Research Achievements

これまでその高い相同性からeIF4A1とeIF4A2には機能的差異が無いと信じられてきたが、本研究によって、その機能には大きな違いがあることが明らかになった。また、eIF4Aは元来、翻訳を促進する因子であるが、本研究によってその前提とは全く逆の翻訳抑制させる機能があることが示された。特に、栄養飢餓などの特定の細胞状態時、特定のmRNAにその機能が発揮されると想定される。以上のような翻訳活性化因子が機能を逆転させる例は統合ストレス応答のeIF2alphaでもみられることから、他の翻訳因子についても同様の機能が備わっている可能性示唆され、学術的な展開が期待される。
