Outline of Annual Research Achievements

本年度、パイエル板内共生細菌群による腸管免疫制御を解明するため、アルカリゲネス菌由来の膜小胞を介した生理活性分子の輸送に着目した解析を進めた。
膜小胞は細菌間情報伝達ツールの一種として知られている一方で、Pseudomonas aeruginosaを始めとした病原性細菌では、膜小胞へ毒素タンパク質や溶血性ホスホリパーゼcなどを封入し標的細胞へ送り込むことが知られている。本年度、アルカリゲネス菌由来生理活性分子の動態に着目し、共焦点顕微ラマン分光計やガスクロマトグラフィー質量分析法などを用いた解析を行った結果、アルカリゲネス菌が膜小胞内に高濃度の短鎖脂肪酸を封入し、放出していることが明らかとなった。
さらに、グラム陰性細菌の膜小胞産生誘導経路として、DNA損傷に伴うSOS responseや、菌密度の増加により誘導されるQuorum sensingが知られているが、これら膜小胞誘導経路によって、アルカリゲネス菌由来膜小胞の内容物組成が変化することを新たに見出した。SOS responseやQuorum sensingは、アルカリゲネス菌を始めとする腸内細菌叢が、宿主の腸管環境下で誘導されることが知られている。そこで今後、腸管組織内部や樹状細胞内部での膜小胞放出やその内容物、樹状細胞の機能制御に着目した解析を進めることで、アルカリゲネス菌由来の膜小胞を介した生理活性分子の輸送や免疫制御能を明らかにしていく。
