Outline of Annual Research Achievements

男性ホルモンであるテストステロンは同化作用を有するステロイドホルモンであり、骨格筋機能において重要な役割を果たす。この血中テストステロン濃度を規定する要因の一つに遺伝子多型が挙げられ、欧米人を対象とした全ゲノム関連解析によってX染色体上に存在するFAM9B遺伝子多型が血中テストステロン濃度に関連することが明らかにされている。そこで、本年度はこのX染色体上に存在するFAM9B rs5934505に着目して検討を進めた。まず、日本人成人男性を対象としてFAM9B rs5934505と握力の関連を検討したところ、FAM9B rs5934505のTアレル保有者において握力が有意に高い値を示した。次に、別の日本人コホートにおいて骨格筋中のmyosin heavy chain isoform(MHC)とFAM9B rs5934505の関連を検討したところ、Tアレル保有者においてMHC-IIxの割合が有意に高い値を示した。さらに、国際大会出場レベルの日本人エリートアスリートにおいてこのFAM9B rs5934505の頻度を検討したところ、Tアレル保有者の割合が持久系アスリート<コントロール群(一般人)<瞬発系アスリートの順に高くなることを確認した。このことから、日本人においてFAM9B rs5934505は骨格筋中の筋線維組成を規定する要因の一つであり、筋線維組成を介してアスリートステータスに関連する可能性が示された。今後はこれまでにアスリートの競技力に関連することが明らかになった遺伝子多型について、どのような機序で競技力に関連するのかを明らかにすることを目標とし、研究を進める予定である。
