Outline of Final Research Achievements |
Previous epidemiological study reported that incidence of pancreatic ductal carcinoma (PDAC) is low in patients who have allergy, in particular received treatment for long time. Thus, we hypothesized that anti-allergy drugs have chemopreventive effect against PDACs. In hamster PDAC model, only Montelukast (Mont), one of leukotriene antagonist, prevented pancreatic carcinogenesis in four drugs. As one mechanism, Mont modulate secretion from pancreatic stellate cells, and it may be involved in down-regulation of proliferation via Smad3 pathway.

Academic Significance and Societal Importance of the Research Achievements |
膵癌は治療抵抗性で、5年生存率の最も悪い固形癌の一つであるため、膵癌になる前予防するという化学予防という考え方も重要になる。さらに、近年ドラッグリポジショニングという考え方は安全性の担保された薬剤を用いるということで臨床応用につながる可能性は高い。その中で疫学調査を元に、ロイコトリエン受容体拮抗薬が膵発癌予防剤として有効であるとin vivoで証明出来たことは、同薬を膵発癌に対するドラッグリポジショニング薬として用いる臨床基盤として示す事が出来たことは意義深く、社会に対する還元力も高い。
