Outline of Final Research Achievements |
The establishment of a disease-specific and effective therapy for lumbar spinal stenosis is a major issue. The purpose of this study was to generate an animal model that can be used to elucidate molecular biological pathology and to determine drug effects in accordance with the pathology of human lumbar spinal stenosis. The animal model in this study is intended to induce degeneration of spine column by resection of intervertebral joint of rats, to chronically develop lumbar spinal stenosis such as hypertrophy of the yellow ligament. The rats were anesthetized, and intervertebral joint was removed to induce instability. In the experiments of Rotarod behavioral analysis, our data suggests that the walking time is shortened by intervertebral joint resection, which is a clinical symptom of intermittent claudication. In the future, it is necessary to examine the degree of thickening of the yellow ligament in histological studies.

Academic Significance and Societal Importance of the Research Achievements |
腰部脊柱管狭窄症の病態は未だ解明されておらず対症療法が主であり薬物無効な場合には手術治療が選択される。しかし高齢ゆえに手術ができない場合もあり、より効果的な疾患特異的な薬物治療確立が大きな課題である。本研究の目的はヒト腰部脊柱管狭窄症の病態に即した動物モデルの確立である。特色は手術でラットの椎間関節切除により脊椎不安定性を惹起し、変形から慢性的に脊柱管狭窄を発症させることである。行動解析の実験では間欠性跛行の指標とするロタロッドで椎間関節切除によりラットの歩行時間が短くなると示唆するデータが得られている。本動物モデルで病態が解明され、新規薬物開発が進められることの社会的意義は非常に大きい。
