Research Abstract

ロジウム(II)-カルボニルイリド複合体を反応性分子素子とする連続型分子骨格構築法の開発および生物活性物質の触媒的不斉合成を検討し、本年度は以下の成果を得た。
(1)分子内にホルミル基を組み込んだα-ジアゾ-β-ケトエステルをカルボニルイリド前駆体、フェニルアセチレン誘導体を求双極子剤とする分子間不斉1,3-双極付加環化反応にRh_2(S-TCPTTL)_4を触媒として用いると、最高97%の不斉収率で付加環化生成物を得ることができた。また生成物の二重結合の立体選択的水素化、ベンゼン環への酸素官能基の導入を経て生薬テイレキシ(〓〓子、Descurainia sophiaの種子)の成分デスクライニンの触媒的不斉合成を行った。
(2)α-ジアゾケトンをイリド前駆体、芳香族アルデヒド求双極子剤とする不斉1,3-双極付加環化反応にRh_2(S-BPTV)_4を触媒に用いると、完壁なジアステレオ選択性および最高94%の不斉収率でエキソの二環性アセタールが得られることを見出した。また、付加環化生成物から5工程の変換を経て抗Helicobacter pylori活性を示す天然物プソラコリフォロール類の触媒的不斉合成を達成した。
(3)分子内にインドールを組み込んだα-ジアゾ-β-ケトエステルを基質とするイミドカルボニルイ・リドの分子内不斉1,3-双極付加環化反応を行うと、完壁なエンド選択性かつ66%の不斉収率で五環性化合物が得られることが分かった。
