Research Abstract

我々はこれまで、ペプチドトランスポータPEPT1の臓器分布、輸送特性、活性調節機構、発現変動機構などについて検討を行うとともに、数理学的モデルに基づくPEPT1輸送シミュレータの開発を進めてきた。しかし、PEPT1の細胞膜局在化機構については未だ不明な点が多い。本研究ではPDZ蛋白質に着目し、PEPT1の細胞膜局在におけるPDZ蛋白質の役割について検討を加えた。PEPT1は肝臓において細胞膜ではなくライソソームにおいて機能している可能性が報告されていることから、まず培養ヒト肝がん細胞HepG2を用いて検討したところ、HepG2細胞にはPEPT1 mRNAの発現が認められたものの、細胞への取り込み活性は全く認められなかった。また、HepG2細胞にEGFP-PEPT1融合蛋白質を発現させたところ、細胞膜ではなくライソソームと考えられる細胞内への局在を示じた。PDZK1またはPDZK2を共発現させてもその局在に変化は認められなかったことから、PDZK1,PDZK2が肝臓におけるPEPT1の局在化には関与していないものと考えられた。次に培養腸上皮細胞Caco-2を用いて検討を行ったところ、Caco-2細胞にはPDZK1,PDZK2が発現していることが示された。そこで、RNA干渉法を用いてPEPT1,PDZK1,PDZK2それぞれをノックダウンしたところ、PEPT1をノックダウンした場合には輸送活性の低下が認められたのに対し、PDZK1,PDZK2をノックダウンしてもPEPT1の活性には変化が認められなかった。従って、PDZK1,PDZK2はPEPT1の局在・活性調節には関与していないことが示唆された。
