Research Abstract

AIRE遺伝子の発現制御下に蛍光分子マーカーGFPを発現するAIRE/GFPノツクインマウス(AIRE/GFP-KI)を作製した。その結果予測通り、AIRE/GFP-KI胸腺の組織切片を抗GFP抗体と抗AIRE抗体によって二重染色したところ、ほとんど全てのGFP陽性細胞はAIRE陽性であった。興味深いことに、AIRE遺伝子をホモで欠損する個体(gfp/gfp)の胸腺でもGFP陽性細胞が同程度に認められ、このことからAIRE自体はAIREを発現する胸腺髄質上皮細胞(mTEC)の系列決定に関与しないことが明らかになった。次いで、AIRE/GFP-KI (+/gfp)およびAIRE/GFP-KI (gfp/gfp)のAIRE発現細胞およびAIRE非発現細胞の各分画をソーティングし、insulin 2およびCRP即の発現をreal-time PCRを用いて解析した。その結果、AIRE-dependent TSA geneであるinsulin2の発現はもっぱらAIRE発現細胞から正常AIRE蛋白質の存在に依存して認められるのに対し、AIRE-independent TSA geneであるCRPの発現はAIRE発現細胞とAIRE非発現細胞の両方から、正常AIRE蛋白質の有無に関わらず検出された。AIREは終末分化段階にあるmTECで発現しているという最近の報告とも併せ、mTECにおけるTSA発現機構として、terminal differentiation model を支持する結果であった。他方、免疫祖織染色からはAIRE/GFP-KI (gfp/gfp)のAIRE発現細胞は形態変化と分布異常を認めたことから、AIREがmTECの細胞構築化に対しても重要なはたらきを持つことが明らかになった。
