Research Abstract

骨髄選択的にプロスタサイクリン受容体欠損させたマウスを利用して、「骨髄のIP欠損は, 血管リモデリング増悪に重要であること」をあきらかにし、「骨髄由来内皮前駆細胞(EPC)の機能維持にプロスタノイドなかでも、プロスタサイクリン(PGI2)系が重要な役割をはたすこと」、さらに、骨髄特異的にプロスタサイクリン受容体(IP)を欠損させたマウス実験動物を用いて、「血管リモデリング、末梢虚血などの病態にEPCのPGI2/IP系が重要であること」を明らかにし、本研究申請目標のほとんどを達成したものと考える。
なお、本研究の成果のうち、(1)血管リモデリングに関する研究は、すでに投稿中である。(2)末梢虚血性疾患におけるEPCに関する研究は、現在投稿準備中である。本研究の成果として重要な点として、強力な抗動脈硬化作用をもつPGI2の、新たな作用機序を明らかにしたこと、ユニークな実験動物モデルの作成により、in vivo各種病態でのEPCの役割、重要度などを明らかにしたこと。また、すでに臨床利用されているIP刺激薬のさらなる応用性についても提唱した点も重要である。
