網膜性色素変性症RP9の原因遺伝子PAP-1の機能と治療薬の確立

Research Abstract

我々はPAP-1の結合因子を質量分析により網羅的同定を行ったところ、これまでに5種類の結合タンパク質を同定した。驚いたことにその中の一つに前述のEPB41L5が同定されたことから、劣性型RPの原因遺伝子Crb1と優性型RPの原因遺伝子PAP-1が共通の結合因子EPB41L5を持つことが明らかとなった。EPB41L5にはC末端が異なるL、Sの2種類のスプライシングアイソフォームが存在するが、PAP-1結合因子として得られたのはC末端側が短いアイソフォーム(EPB41L5-S)だけであった。また、EPB41L5-Sは通常細胞質に局在するが、PAP-1の存在下では核内に蓄積することを観察しており、PAP-1はEPB41L5-Sを核内にリクルートできると考えている。
これらの知見は、劣勢、優勢の網膜色素変性症に共通な分子基盤が存在することを意味しているのかもしれない。
更に、PAP-1のスプライシングターゲット遺伝子の探索を行い、複数の候補遺伝子を同定した。
