Research Abstract

昨年度に引き続き,コオロギ胚発生において前後軸パターン形成に関与する遺伝子の発現パターンおよび機能の解析を行った.ショウジョウバエと異なる細胞環境における転写因子の分布パターンを明らかにするため,caudal(cad)やorthodenticle(otd)等の主要な遺伝子の胞胚期での発現パターンを解析するためのwhole-mount in situ hybridizationの技術を確立することに成功した.この技術を用い,otd遺伝子が初期胞胚期には比較的広範囲に発現し,やがて形成途中の胚原基の前方に発現が限局することを明らかにした.このことは胚原基が形成された段階で,すでに頭部の領域が決定されていることを示唆する.cad等他の遺伝子についてもデータを得ている.この成果について,日本発生生物学会,日本分子生物学会において発表し,現在論文を準備中である.
一方,ショウジョウバエの脚のパターン形成に関与することが知られているextradenticle(exd),homothorax(hth)遺伝子について,RNAiによる機能解析を行ったところ,これらの遺伝子が,コオロギで体節形成に関与していることが明らかになった.特にexd遺伝子についてはギャップ遺伝子のレベルで働いていることを示した.この新規の知見はDevelopmental Biology誌に掲載された.
