Research Abstract

ピロリジン型イミノ糖が活性部位指向的なケミカルシャペロン効果を持つことを見いだした。これら化合物は、細胞表面の糖鎖構造に影響を与えることなく、Gaucher 病モデル細胞内のβ-glucocerebrosidase活性を増加させた。これまでピロリジン型イミノ糖は、水溶液中におけるコンフォメーションが安定せず、特定のグリコシダーゼに対し選択的に結合することは難しいとされてきた。本研究成果により、特定の誘導体化により生体内のpHに依存することなく、ほぼ一定のコンフォメーションを保たせることが可能であることを明らかにした。従って、これらピロリジン型イミノ糖は、新しいタイプのケミカルシャペロンと成り得る可能性が期待される。
