Outline of Annual Research Achievements |
In FY2019, the fellow’s role was to develop the experimental model to test synthetic regulatory circuits using transcription activator-like (TALE) proteins fused to activator or repressor domains (Crocker et al., 2016). The amount of the TFs contribution is manipulated by adding or subtracting the number of TALE-binding sites in the enhancer. The same approach with TALE fused to Bcd was applied; by adjusting the number of TALE binding sites, the impact of the stepwise increase in Bcd activity will be observed, so testing whether and at what point it switches from repressor to activator. Experimental conditions for generating and assembling essential fragments that compose of transgenic constructs were optimized. They can then be used to create transgenic flies for applications later on. Altogether, the success of these steps provides a strong technical basis and shows great promises for downstream experiments.
