Outline of Annual Research Achievements

本研究では、ウリ類炭疽病菌の宿主キュウリへの侵入ステージにおいて高発現するエフェクター群に焦点を当てる。ウリ類炭疽病菌の6系統のいずれにおいてもキュウリ侵入ステージにおいて高発現しているエフェクター様分泌タンパク質遺伝子群を網羅的に選抜し、続いて標的破壊解析を完了した結果、ウリ科作物への病原性に関わる4種のエフェクター遺伝子、EPC1、EPC2、EPC3 (旧名ECAP12)、EPC4の同定に成功している。本年度は、このEPCエフェクター遺伝子の発現パターンを明らかにした。また、EPC1、EPC2、EPC3、EPC4遺伝子に対する4重遺伝子破壊株をCre-loxPシステムによる多重遺伝子破壊方法により作出した。その結果、4重遺伝子破壊株はウリ科作物に対する病原性はほぼ失う一方、ベンサミアナタバコに対する病原性は本菌野生株と同様であった。本結果より、発見に成功した4種のエフェクターEPC1、EPC2、EPC3、EPC4は、ウリ類炭疽病菌のウリ科作物に対する宿主特異性成立において重要な役割を担っていることが強く示唆された。また、EPC3 を過剰発現させたベンサミアナタバコについて、PAMP誘導免疫の状態を調べた結果、細菌PAMP(flg22)によって引き起こされる活性酸素生成をEPC3は顕著に抑制することを明らかにした。さらに、EPC3を恒常的に発現させる形質転換シロイヌナズナを作出し、同様にPAMP誘導免疫への影響を調査した結果、EPC3はシロイヌナズナにおいても、flg22誘導活性酸素生成を明確に抑制した。
