Outline of Annual Research Achievements

AMOR活性を担う2糖構造は、トレニアの雌しべに限らず、広く様々な植物、そして様々な器官のAG糖鎖に見られる末端構造である。しかし、雌しべの中でもその分布は一様ではないように(Mizukami et al., Curr. Biol. 2016)、特定の発生ステージや、植物体の特定の位置で、様々な細胞間コミュニケーションを担っている可能性がある。

研究計画1(AMORの新たな機能の探索):AMOR生合成系に関して、トレニアにおいて見出したグルクロン酸転移酵素やメチル基転移酵素の候補について、準備してきた、ゲノム編集による遺伝子破壊株の解析を引き続き進めた。昨年度までにトレニアにおける遺伝子群のリスト化を終え、投稿準備中のゲノム論文に含める準備を進めた。表現型を解析することで、AMORの新たな機能を探るために、野生型へのバッククロスにより、培養変異の除去を進めた。樹立に成功したモノクローナル抗体を用いて、AMORの分布およびこれらの変異体での変化を探ることを試みた。モノクローナル抗体を用いた免疫染色を進めた。これにより、初めて胚珠でのAMORの分布が明らかとなった。AMORは、ヤリブ染色でAGPsが多いことが示唆されていた部位に分布した。

研究計画2(AMORの受容メカニズムの解析):AMORが発見されたトレニアのAMORアッセイ系を用いて、花粉管におけるAMORの受容メカニズムを探った。トレニア近縁種であるウリクサにおいても、AMORアッセイが容易であり、同様にAMORが機能することが明らかになった。ウリクサを遺伝学が可能なトレニアとして基盤整備を進めることができたことは、今後のAMOR研究の発展のために非常に重要な展開である。有機化学合成により改変した多量体AMORなどを使用し、可視化や相互作用分子の同定を達成することで、AMORの受容メカニズムを明らかにする基盤も確立できた。
